TritonX 100 is not removed by a dialysis method
Affinity purified proteins are dialyzed with the optimal buffer prior to using for the enzymatic activity. Although we usually use Triton X100 as a detergent to lyse the cellular structure to purify target proteins, the detergent has a possibility to negatively influence to the enzymatic activity. Therefore, it is important to remove the excessive amount of Triton X100 from the buffer including target proteins. However, the dialysis method using membranes with tiny pores is not suitable for this purpose, because Triton X100 composes micells that makes it difficult to penetrate the membrane during the dialysis procedure. Accordingly, if we plan to remove Triton X100 from buffers, we should consider the other method; for example, selecting a washing buffer for affinity column without much amount of Triton X 100.
