I have many unpleasant memories regarding an experiment, immunoprecipitation. Many careful checkpoints are needed for achieving this experiment perfectly. Once, I had performed this assay without a negative control, normal IgG, unbelievable! I used to wonder whether the appearing band by my assay is true or not, because of the lack of a negative control, which I can consider now. I should have asked some senior researchers about the experiment and my trouble. Someone should have told me that I must know the fundamental skill of the assay.
Many shameful mistakes made me a better researcher, which resulted in the growth of my experiment skills. Now I am able to try an immunoprecipitation with minimum procedures, without a terrible mistake. Even currently, it is sometimes difficult to obtain a good performance of immunoprecipitation, maybe due to the characteristic of the objective protein and quantity of an antibody against it.
