The efficacy of one- and two-doses administration of adenovector vaccine encoding Wuhan type SARS-CoV-2 S gene was evaluated by using adult and aged non-human primates (rhesus macaques). The two-dose vaccination more profoundly induced higher binding and neutralizing antibody responses than the one-dose vaccination. The one-dose vaccination enoughly induced humoral immunity and T helper cell-dependent cellular responses in both adult and aged animals, which protected animals from challenge of SARS-CoV-2 D614G mutant. Neutralization of sera from these vaccinated animals was efficient for several variants including B.1.1.7 (UK. variant) but not for B.1.351 (S. Africa variant).
